The easiest answer for anything like this in RE is because the plot needs it to happen.

The guy is the village guard. His job is to tell people to GTFO, and if they don't his buddies down the path make sure they have an "accident".
Remember also that the plagas communicate on sound frequency or something we can't hear, so when his plagas whispers to the other "some American just showed up asking about the girl", who ever was leading probably said, "oh snap, drop the bridge and kill that guy." until Mendez was able to catch up and apprehend Leon himself.
Remember, the village has an active interest in NOT getting a bunch of attention because they're planning a coup, so they are trying to avoid doing anything which brings in more cops/soldiers/agents. Leon showing up is basically the signal that some governments know the US President's daughter is there, so the facade drops and everyone goes in to "murder outsiders" mode.

Because he was just a regular homeowner trying to ward off and then defend himself from an armed American intruded randomly entering his house talking in a foreign language. Once Leon kills him the rest of the village hears that an armed lunatic has gunned down one of their own and tries to fight him off to defend themselves.
